SLC38A4 expression was confirmed by western blot in multiple tumor cell lines. In mouse models, liver tissue imaging (H&E staining) showed that overexpression of SLC38A4 reduced metastatic tumor areas, while knockdown increased them across Hepa1-6, MC38, and B16F10 cells. Results were consistent in both immunocompetent and nude mice, with statistically significant differences.

Longevity is defined by lifespan and factors affecting it so in this blog we are going to know about some suprising facts about the longest living animals and the reason behind their high life span , mechanism behind it and many more . But before that we will discuss about what is longevity , what are the factors affecting it and what is the mechanism.

Longevity

It is defined by the ability to live a long life beyond the species-specific average age at death or beyond the original life span or in short longevity means long life . Some organisms are able to live longer than their original life span which makes it intresting to know the reasons behind longevity .

Factors Influencing Longevity

The factors which influences longevity are as follows :
  • Slow Growth Rates : It Lowers the metabolic rates which leads to reduction in cellular stress thus increasing life span.
  • Low Metabolic Rates : It efficiently use the energy minimising the oxidative damage resulting in long life .
  • Adaptation to Stable Environments : Adaptions help in resisting the harsh environmental conditions by adapting some specific features which allow them to survive there thus reducing stress from environmental fluctuations or environmental changes.
  • Ability to Repair or Regenerate Tissues : Maintaining cellular integrity by rapid regeneration and repair does not allow the death of cells or tissues which means the death of the organism also get delayed.
  • Genetic Predisposition : Nature is the best creator which choose wisely the fittest and best organisms to live longer through Evolutionary selection like natural selection and survival of the fittest.
  • Diet and Nutrition : Nutrient-rich diets or foods supports an overall good health free of diseases which helps in longevity .
  • Lifestyle : Sedentary or slow-paced lifestyles reduces cellular stress leading to a long Life .

Mechanism Behind Longevity:

  • Telomere Maintenance :

Telomeres are the Protective caps on the ends of chromosome and shortening of which causes aging and cellular senescence and this can be prevented by “ Telomerase “ the Enzyme responsible for maintaining telomere length thus increasing life span slowing the senescence.

Examples: Telomerase-active cells such as stem cells resist aging

  • Epigenetic Regulation:

Epigenetics is the Gene expression which is controlled through DNA modifications by modifying the Histone protein which is responsible for regulation of Chromatin structure . By DNA methylation through Gene silencing and activation it will help to increase life span.

Examples : Epigenetic changes influence aging and development

  • Stem Cell Maintenance:

Stem cells are Self-renewing , differentiating cells which Contributes to aging and tissue degeneration through stem cell exhaustion . Stem cell niches help in supporting in Microenvironments . Examples: Stem cell therapies aim to restore tissue function

  • Antioxidant Defences: Oxidative stress bringsl out Reactive oxygen species (ROS) which is very harmful to the cells, many antioxidants have been reported to help in the inactivation of ROS and thus having the cells in good conditions.

Examples: Vitamins C and E, beta-carotene, and polyphenols are some examples while Antioxidant enzymes involve Superoxide dismutase, catalase, and glutathione peroxidase.

Autophagy:

Autophagy is the cellular self-cleaning in which hydrolytic enzymes destroy the damaged cells and recycling of damaged components occur. Thus Autophagy-related genes are very Essential for longevity and health and Autophagy-inducing compounds like Potential therapeutic strategies helps in this process.

Examples: Autophagy-deficient mice show accelerated aging

  • Mitochondrial Function:

Mitochondria is the energy producing organelles also called power house of the cell hence Mitochondrial dysfunction will cause aging and disease . For longevity Mitochondrial biogenesis is required as it Enhances energy production thus leading to longevity .

Examples : Regular exercise and caloric restriction food intake promote mitochondrial function

  • Hormonal Regulation:

Hormones are the Signalling molecules influencing aging and longevity while Insulin/IGF-1 signaling Regulates growth, metabolism, and longevity and responsible for hormone-related diseases like Diabetes, cancer, and osteoporosis etc

Examples: Hormonal therapies aim to promote healthy aging and longevity

  • Genetic Stability:

DNA repair helps in Maintaining genome integrity while genome instability can leads to aging and cancer . Genetic stability mechanisms such as Telomere maintenance, epigenetic regulation etc helps in longevity.

Examples: DNA repair-deficient mice show accelerated aging

Top 5 longest living animals till date are as follows:

Here are the top 5 longest-living animals recorded till date are as follows:
  • Turritopsis dohrnii : Also known as Immortal Jellyfish which has an estimated lifespan of 12,000 years according to Scientific record . It lives in Tropical and temperate waters . It’s Unique feature which helps it to live for these long years is Transdifferentiation which allow it to revert to polyp stage whenever necessary.
  • Ocean quahog or Arctica islandica have an estimated lifespan of Up to 562 years which is verified by scientific records . It is found in North Atlantic Ocean and Slow growth rate that is about 1-2 cm/year is it’s unique feature which helps it for longevity.
  • Bowhead whale or Balaena mysticetus have an Estimated lifespan of Up to 245 years but according to Scientific record it is 211 years . It’s habitat is Arctic waters and Unique feature is Slow growth rate, about 1-2 cm/year
  • Koi Fish or Cyprinus carpio has an estimated lifespan of 226 years and it’s habitat is Freshwater ponds and lakes Unique feature is Slow growth rate, about 1-3 cm/year
  • Rockfish or Sebastes spp has estimated lifespan of 205 years and it is found in Deep-sea environments while it’s Unique feature is Slow growth rate, about 1-5 cm/year

Other than these the other long living animals are

  • Tortoises ( up to 188 years )

Galapagos Giant Tortoises

Aldabra Tortoises

  • Tuataras (up to 177 years)

It is native to New Zealand and Considered as living fossils .

  • Whales (up to 110-150 years)

– Bowhead whales

– Sperm whales

– Fin whales

  • Corals (up to 400-500 years for some species)

– Brain corals

– Reef-building corals

  • Sponges (up to 2,300 years)

– Some species of deep-sea sponges

Interesting Facts :

  • Some species of rocks and corals can live indefinitely through colonial reproduction.
  • The longest-lived human on record was Jeanne Calment , who lived for 122 years.
  • Some animals, like certain species of jellyfish and flatworms , are biologically immortal because they uses trans differentiation to replenish the cells after sexual reproduction .

Longevity-Associated Genes Involves :

  • SIRT1 : This Regulates metabolism , stress resistance, and longevity
  • FOXO : Transcription factors which influences longevity and stress resistance .
  • p53 : Tumor suppressor gene which regulates cellular senescence enhancing longevity .
